Cyano-acrylate superglues are anaerobic adhesives. As long as there is air in the tube they do not polymerise. As soon as air is excluded (by pressing together the surfaces to be joined) the glue polymerizes.

From the Loctite website:
Q: Why does Loctite only fill half of their anaerobic 50 ml and 250 ml bottles?
A: Actually, there is exactly 50 ml and 250-ml adhesive in each package. We fill bottles halfway because air is needed in the package to prevent our anaerobics from solidifying. Our 50-ml and 250 ml bottles also allow our anaerobics to breathe by allowing air to permeate through the bottles.

If it went hard in the tube you would not be able to use superglue so the companies who make it make sure that they make it so that it does not go hard in the tube.

The substance has to react with the moisture in the air to harden - there's no air or moisture in the tube so it doesn't harden.

super glue reacts when it comes into contact with water vapour in the air... it was infact designed as a sort of temporary "stitch" for deep flesh wounds.
as there is no air and therefore no watervapour in the tube it does not react and become hard.

I think it's to do with the chemcial reaction to the air. As long as it's in the tube with no air getting to it, it's fine, but as soon as you squeeze some out and it gets into the air , then a checmical reaction takes place which makes it set hard.
