Centerfire rifles have a round bolt and the firing pin comes out of the middle of the bolt face and hits the center of the round.... most rifles above .25 caliber are center fire.....
Rimfire.... rifles and handguns have a striker that is square shaped and when you pull the trigger, it hits the edge or rim of the round and thats how it fires........ most hand guns and rifles .25 and under are are rimfire......

There is also the power factor in the difference between the two types of rifles.
The rim fire rifles are very light and made only for target shooting and small game.
The center fire rifles are bigger and made for big game, varmints and anything on the planet. They are also heavier and have more recoil.
Expense is another difference, the center fire rifles cost more and the ammunition costs more too.

Rimfire ammo had the primer compound around the rim of the catridge, centerfire ammo has a small or large primer, and the brass is reloadable, whereas the rimfire is NOT reloadable.

The bullet for a center fire rifle has the primer located in the center of the base of the bullet and the firing pin hits directly on the primer. Center fire shells can be easily re-loaded if you get into that. Rimfire rifles are smaller in caliber .22 or .17 and the firing pin hits the rim of the shell to ignite the primer. To my knowledge you can not re-load rim fire shells.
