As you can imagine from my name I love New York and I have been 5 times 6th time will be in Dec.
You have to go to Top of the Rock @ Rockerfeller Centre the views are far more fantastic than from the Empire State Building. The ESB is great too but at the Rock you actually get the ESB in your photo's.
Go to the South Street Seaport.
www.southstreetseaport.com
this is great for shopping, eating and magnificent views of the Brooklyn Bridge.
Speaking of Brooklyn Bridge you must walk over it to Brooklyn. It's a great walk, with great views of Manhattan, Brooklyn and also Statue of Liberty.
Take the Staten Island Ferry as it is a free way to see the Statue of Liberty and get great photo's.
Obviously visit Times Square and the Theatre District, Radio City Music Hall and Central park. 5th Ave for shopping. If you are there in Jan/Feb you can go Ice Skating in Central Park Woolman Rink or Rockerfeller Centre.
Visit Macy's and Herald Sqaure. Also Riverside Park is beautiful.
Get a New York Pass.
www.newyorkpass.com
it will save you loads of money, I can recommend it we always get one.
Also get a Subway Pass for 7 days, it cost $24 and you can use it all week. Much cheaper than Cabs.
GrayLine Bus tours are great too.
You will find so much to do it is a fantastic city. Enjoy

If you have not been before and may not go again for a long time than the must see things that are free or nearly so are:
Central Park
The Statue of Liberty
Times Square
Rockefeller Center
Shopping in Soho/Grenwich Village
View from the Top of the Rock
However, it also depends on your budget since New York has some of the best hotels, restaurants and night clubs in the world. Not just high end, but eclectic hotels eateries and clubs. There is literally something for all tastes.
In addition to the must sees listed above I would suggest seeing a Broadway show. The most popular one in town now is Clueless, but there are many great shows and if you budget is tight than go see a matinee or an off Broadway show.
If you want to get eh most out of Your New York City trip than spend some time planning it. Only you know what kind of vibe you like and your favorite food. You can get the best of what you want if you spend a little time preparing. I listed a couple of sites in the resources that have videos and photos of about everything in New York. You can also find deals on hotels and shows. Anyway, a pictures worth a thousand words so check out some popular NYC web sites.

After the Standard stuff (Statue of Liberty, Ground Zero, Empire State Bld, Chinatown, Little Italy, The Metropolitan Museum etc.) It comes down to what you enjoy doing.
Clubs, Biking in Central Park, shopping, Music, Architecture, history, etc. Each of these subjects (lack of a better word) can be made a tour in NYC. Also you can explore NYC by it's neighborhoods. Lots of different ways to go at it. Are you a 'Night Owl', a party animal, a picture taker, a history buff etc ?
There are many books and on-line sites regarding NYC touring, more important is 'What rings your bell'.
One item - if you get homesick check out the below Website - have a pint with a countryman while in Greenwich Village - Little Britain

If you go down to the WTC site, go across the street to St. Paul's - they have a display of pictures, etc. from 9/11. The WTC site itself looks more-or-less like a construction site....
Also - take the [free] Staten Island Ferry for a great view of the NY skyline and the Statue of Liberty.
