Since Katrina, Amtrak haven't been running from Florida to Louisiana. The Sunset Limited currently runs from Los Angeles to New Orleans only.

If you want to do the whole trip by train, the only way is to catch a Silver Service train up to Washington, DC, the Capitol Limited or Cardinal to Chicago, and then the Texas Eagle to wherever you're going.

Or you could catch the Crescent from Washington, DC to New Orleans and then the Sunset from there.

It would be far quicker and cheaper to rent a car.
Just head east on Interstate 10 then, depending on where in Florida you want to go, south on I75 or I95.
