buy compatible components and software (namely an operating system) and plug and pray...err i mean plug n play.
you'll need:
Case
Power supply (comes with some cases)
Motherboard
CPU
Heatsink&fan for CPU
RAM
Hard Drive
floppy drives (optional)
Graphics card (optional if motherboard has an integrated graphics card)
sound card (optional if motherboard has integrated sound)
network card or "NIC" (network interface card) (optional if motherboard has integrated NIC)
optical disk drive (CD/DVD ROM or RW)
speakers (optional, but...come on!)
keyboard
mouse (optional, yes I said the mouse is optional...viva la command-line!)
monitor
wear an anti-static bracelet clipped to the case while you plug it all together, read the appropiate manuals to make sure everything plugs in to the right spots. everything should come with enough cabels and wires to connect everything.
Or if you want a more simplified, but not a do-it-yourself project...

I've built many PCs from 'scratch' over the years.
I always start at http://www.pricewatch.com
They list components by type: cpus, motherboards, cases, io cards, memory, disk drives, software, etc. You can start with a bare-bones system, or build from components.
It is always best to start by choosing a motherboard. The motherboard will dictate the type and speed of the cpu, the type of memory, the size of the case and power supply, type of disk drive, etc. Then buy the rest of the components to fit the motherboard.

PC magazine publishes regularly on it. Recently someone built the best system you can get for $1,900 and explained every single component of it. (they didn't actually explain how to wire everything together though) See: www.pcmag.com
