If you MUST travel direct...take the Indian Pacific train.
It'll take longer, allow you more time to relax, and in a sleeper cabin...is very comfortable.
If time is of the essence, hop a flight.
I seriously doubt that there'll be a direct flight to Kalgoorlie from Melbourne...but it's easy enough to get to Perth and it's then only another 90 min flight from there to Kal. Alternatively, you could also train or bus from Perth up to Kal...takes about 8hrs though.
Costs for either option will be about the same.
The only other direct route will be by bus.

If you can afford it and have the time, take the train. It is a wonderful trip and you can explore Adelaide on the way. Get a sleeper, don't even think about sitting up. If time or price is important, fly. You might find it cheaper to fly to Perth and then take a bus or train to Kalgoorlie.

Everything I've heard indicates that it isn't any cheaper and takes much longer to go by train. However, if you have the time, and can afford a sleeper car...then by all means.

Fly mel to perth and then train (prospector) or bus to kalgoolrie generally this is the cheapest way.
Rail although more direct is generally more expensive when you take into account time and meals on board the service.
