V6 is a 6 cylinder engine in which 3 cylinders are placed on each side of the block; creating the V shape (thus making it a V6) It is possible to find an inline 6 cylinder, but not likely.

The six refers to the number of cylinders in the engine. The V refers to the cylinders being configured in a V pattern with three cylinders in each bank meeting at the bottom where they join at the crankshaft to create a V.
There are a number of other common configurations some of which you have probably heard of as well.
V-8--Two banks of 4 cylinders each meetng at the crank.
Straight Six--Six cylinders in line with the crank at the bottom.
Horizontally Opposed Four (also called a waterboxer if water cooled)--In this case you have two cylinders on each side of the crank laid out in a flat pancake design. Old air-cooled VW beetles and some small Porsches had this. Later VW Vanagons had the waterboxer, as do most current Subarus, although the larger Tribeca SUV has a horizontally opposed six with three cylinders per side. The higher end Porsches also have horizonally opposed sixes.
V-10--Five cylinders per bank in a V pattern joined at the crank, found in some Dodge trucks and the Dodge Viper.
V-12--Found in some Ferraris and other exotics, two banks of six cylinders each joined at the crank.
W-12--Essentially two V-6s welded together with four banks of three cylinders each, found in Bentleys and a few other exotics.
Straight Five--Five cylinders in line found in some Audis, Mercedes, and at one time in the Acura Vigor.
V-4--Rare these days, but some old Saabs had them, two banks of two cylnders each joined at the crank.
Straight Three--One In-line bank of three cylinders found in some of the small Geo Metros and the like.
Slant six--Essentially a straight six turned at an angle to allow for a lowering of the hood-line.
Straight Eight--Eight cylinders in line as found in some of the old Packards, and perhaps others of which I am unaware.
I won't even get into the radial design engines which are mostly used in aircraft, but essentially have however many cylinders arrayed in a circle around the crank.
Then there are the wankel or rotary engines, which these days I believe are the exclusive product of Mazda--triangular rotors orbit inside a sort of figure 8 shaped combustion chamber. I think the first commercially available wankel appeared in the old NSU. Mercedes-Benz also experimented with it in the C-111 prototype but never marketed it.
There are also a variety of configurations peculiar to motorcycles that I will not go into.
I have also left out multiple cylinder marine engines, and auto engines over V-12, as one sometimes saw in high-end cars the 1930s.

It means the motor is V shaped, and it has 6 cylinders.
There used to be straight engines with 6 cylinders, and that would be called a L6

It means 6 cylanders arranged in a v, with 3 on one side and three on the other, they also make a stright 6 cyl. with all of the cyl. arranged in a straight line and a H 6 cyl motor. Imangine taking the V 6 cyl. and bending the V down to a straight line with 3 cyls. on one side and 3 on the other.

That's the configuration of the engine block. Looks like a "V" from the front.
They also have "in line 6", "slant 6", and "opposing 6"
