It looks just like marble so I want the grout line as small as possible or non-existent.

Is there a down side to not having a grout line?

I have some 1/8" but.. I might prefer zero if it would likely look good and work out.

u can go with 1/16 just be sure to use "unsanded grout",....

The type of tile has absolutely no relevance. A spacer for a grout line is standard procedure. 12 inch is a rather large back splash; but no reason why you can't use what you want.
