The space between the 2 X4 's is the "rough opening" which should be a half inch wider and taller than the actual door and frame dimensions to allow for shimming to get the door plumb and level. So if you have a 30" door with a 1 1/2 " frame thickness for both sides, then you'll need a rough opening of 32 inches wide. Measure the actual height of the door and frame thickness to get a dimension for the rough opening height allowing for a gap at the bottom of the door for ventilation or to sweep over carpet.
