I have my 456 visa (short-stay business visa) and I am going for a job interview in Sydney, there is a high probability that i will get the job so can i buy a one-way airline ticket to Australia on this visa?

The 456 does not allow you to work, only go to a business conference, job interview etc. Plus it is only good for three months so I'm sure customs will want to see a return ticket. Even if you get the job you will need to go back to your home country and apply for an e457 work visa. It will take immigration 4-6 weeks to process plus the time it takes beforehand to get all you paperwork in order like getting ref's, degree, passport notarized, etc.

I flew here with a 457 using a one way ticket as I didn't know exactly when I would go back. Customs never questioned it.

that visa will not permit you to work in australia and i think you will find you need to prove you have a return ticket before being allowed to enter australia

there is no "best month" - subscribe to receive emails that advise wen specials are on. cheapest tickets are usually the red eye flights. also check how many stops they make en route. sometimes cheapest turns out to be the longest or worst times of the day/night.

buy a two way and just cancel the return if you get the job

I don't see why not. I would call a travel agent just to be sure.
