Yes. How would you define light if you do not compare it against dark? Or definine Good without considering Bad/ evil,etc?
Words are created by us humans, and the english words have so many different meanings, but when we translate other languages into english, for example greek, it is easy to see why it is hard for us to even DEFINE what A is.

It's difficult to define anything with out a reference point. For example, I could say "he is tall". But if he is the only person I've ever met, how would I know what tall was compared to anything or anyone else? Same with a color. One could say "that is purple"...then, could hold it up to a blue or another shade of purple and it could look more blue.
With out a reference point, all descriptions of things are relative. So, I believe that in most cases what defines A is how it is related to or compared to B..or C...or D...
One person's tall could be another person's average and one person's shade of blue could be another's purple.

It is the matter of A that is formed to the letter A. A is distinct from B for it has also a matter of its own to form the letter B.

If A and B are of the same category.......yes.
Eg, with human...a male is a male because he's got penis and no breast (compare with female)
If A and B are of totally different category....no.
Examples here are many. Like comparing orange and apples. Why bother doing it?

yes. in a world where everything is a competition/race, we cannot live without making a comparison. A never really has its own characterictics, but more often than not it is how much distinction can be made from B A is.
