in 45 minutes? Is 5.5 mph a good speed?

Your heart rate and breathing are what determine the ideal effort to burn fat.

You should be going as fast as you can, but still be comfortable. You should be able to breath easily, even hold a conversation, you should not be panting or feel out of breath.

Speed is irrelevant, what is important is your heart rate. Try to keep your heart rate at 70-80% of your maximum heart rate. Most treadmills have a chart telling you what your max heart rate is.

For an extra fat burning boost, do interval sprints on a treadmill.

Stretch
Walk for 5 mins. to warm up.
Sprint a .25 mile.
Walk or Jog .75 mile
(alternate sprint and walk for 20-30 mins.)
Walk for 5 mins. to cool down.

During the sprints, don't jog... really sprint. You will defeat the whole purpose if you are not running as fast as you can. The constant stress / rest alternation is very good for lowering body fat.

Most machines have a meter on the side that says based on your age where your heart rate needs to be, to be burning fat. Plus, each machine is different. Look on the web to see if you can find something that you can customize to your age. It is hard to maintain that range because the tendency is to go faster which is cardio. You can tell it is working because you'll feel your body getting hot. It is pretty cool.

mph depends on the person, whatever speed is a comfortable to maintain, but you feel tired after is what you should do. the harder you go the more your body is working and the more fat you burn, but you will still burn fat at a lower intensity,
