I Have been in Law Enforcement for over 12 years and I have experience in dealing with liars, thieves and con men. The eyes as the others have pointed out are key indicators but there is more to it.
1.Perspiration - Sweating is a bad sign.
2.Gulping - When a person gulps before, during or after the questions asked.
3.Movement - restlessness and fidgeting during questioning
4. Covering - If the person puts their hand to their mouth while answering questions they are subconciously covering up a lie.
5.Lip Biting
6.Re-direction - If the person being questioned tries to distract you or is being extremely helpful in accusing other people and offering suggestions as to how the incident could have occured, anything to take the focus off of them
7.Parotting - Repeating questions directed towards them because they are stalling for time and carefully thinking about, memorizing and reviewing their answers
8.Questioning the Interrogator - Example; the person being questioned asks the interrogator "If I Killed Nicole and that other guy then why would I blah blah blah....and would I have done this if this had happened, what's my motive why would I do that?" etc. etc.
9. Complicated story lines - If someone being questioned gives you the most complicated, detailed, laundry list of things that they were doing on the date of the incident. Talking about anything from their sick Grandmother to their long lost cat Mr. Fluffer Nutter is a sure sign.
10. Defensive - When the accused becomes extremely aggitated and unreasonably irrational. Cursing, throwing a fit or temper tantrum, and finally storming out...that is a defensive mechanism (the fight or flee response).

I know when someone is lying usually by the feeling I get in my stomach. Instincts, not to mention when something doesn't sound right or doesn't fit that is probably because it is not the truth. People will forget their own lies, or what ever they are lying about doesn't make sense to me, or is questionable, and anytime something is questionable I make a decision not to trust the info I have been given, and I inquire more to see if the truth adds up or not

They get nervous and look away. But, only if you ask them something and they act this way. If there confident and look at you in the eye without being awkward, there being honest. Also, ask them why and there answers should not add up. Trick them to find out if there really good at lying.

Look at there eyes. If you ask them a question and they look up to the left they are lying, if the look up to the right they are telling the truth. It has to do with one side of the brain is for memory and the other is for creativity.

Sometimes you can't. But, watch them closely. Twitching, touching their face too often, looking to the left for creativity. All factors of lying.

If they cant look you in the eyes they are hiding something OR if they can look at you but turn away often and look to the right side then they are not telling you the whole truth

They are lying when they cannot look you in the eye and keep their eyes fixed on yours without looking to the side while telling you their delightful fib/exaggeration!!

Their eyes always give them away for me the awkwardness when they speak searching for words. That sort of thing makes it very noticable for me. That and silence is always a dead give away!
