You need to give more information to get an accurate answers Starting and destination cross streets, and times of day would allow us to give you a fairly accurate answer. Just saying "Glendale" and "Downtown" covers too much area.

Thank you for the additional info.
If this fits into your schedule try the Burbank Bus "Empire Line" from Glenoaks / Cohasset to the Burbank Metrolink Station. It operates every 20 minutes 5:30-10:00 am and 2:30-7:30 pm. Fare is $1.00
Transfer to the Metro #92 bus at the Burbank Metrolink Station to (roughly) Olympic and Main Operates every 20 minutes from 6:27 am - 7:18 pm. Fare is $1.25

Total trip time is about 1:45, and this routing saves you $0.50 round trip daily over using only Metro buses.

Could take a flight then rent a car, or you could take public transportation except when you want to make some excursions outside the city on your own, independent of of all the bus tours. Otherwise, it's not so much the traffic as the parking in the city, especially San Francisco. I live north of San Francisco, where the buses aren't quite as frequent as you might like. In fact, the buses might be seen as a time-consuming hassle. So, if you want to see all the scenic back roads, you might enjoy a rental car, just not in the city. Do some research on the cost of rental cars & consider any discounts, such as AAA or Costco, that you might have on them.

Train will take you from Burbank Airport right into Union Station/ Olvera St.
Bureaucracy keeps L.A. from modernizing its public transportation system. And then again the mayor might try to keep his fingers out of the nookie jar.

here's what the Metro trip planner suggested
Metro Bus 292 (BURBANK STATION) heading south
From: GLENOAKS/COHASSET(SW corner) Lv: 10:22AM
To: 1ST ST/OLIVE AV(SW corner) Ar: 10:33AM
Pay $1.25, Monthly Pass: $62.00, (EZ Pass accepted)
Ride Metro Bus 92 ( DOWNTOWN LA - MAIN-OLYMPIC) heading north
From: 1ST/OLIVE(SE corner) Lv: 10:40AM
To: OLYMPIC BL/MAIN ST(SE corner) Ar: 11:46AM
Pay $1.25, Monthly Pass: $62.00, (EZ Pass accepted)
as an alternate you could take the 292 to the Burbank metrolink station and board the metrolink to Los Angeles
then hop on the Dash D bus (which operates via Spring towards the fashion district)
