Do you give them a gift to make them feel appreciated or would a sincere 'thank you from the bottom of my heart' be enough?

Fresa,
depending upon what the favor was (as you know it to be and the relevant importance it holds to you) I would at least consider a card.

In the card provide a hand written note of thanks, expressing in words what it meant to you. Additionally, I would suggest that when the person opens the card they only see the pre-printed words "Thank You". Your hand written expression will be much more appreciated.

Hope this helps and best to you!

Both would be nice but there might come a time when that person needs a favor from you. Let that person know that if they ever need anything that you can do for them, that you will do your best to do so. Although you might not be able to express it yet, one day you might and when that time comes make sure you do all you can to help that person out. It can be a week from now or years from now. I am sure that person will appreciate the return gesture of sincerity.

a big thank you from the bottom of yourheart is always good and should be enough but if spending, a little money won't hurt you a small little token would be in order if you can afford it like a gift card with thank you card saying why you appreciated the favor

If you can afford a gift good otherwise a big thank you will be fine but say it in such a way that will tell her that you really mean it from the bottom of your heart.

If the favor was that big, and you are in a position to afford it, I would either treat them to a nice lunch or dinner, or send them a fruit basket or a cake. Either way, you should include a thank you note or card.

If someone did a really big favor for me, I would return a big favor to them like offer to babysit their children (if any), so they would have a day off for themselves or offer to help clean their house, help cook meals, or whatever help they need, that always doesn't have monetary value.
