First, you'd say "Let's" not "Lets" because it's a contraction of "Let" and "us."

Second, those two sentences mean very different things. The first declares a fixed time for both lunch and the meeting. Lunch, we can assume, is just beginning or is already happening, and the meeting will occur 15 minutes from now, no matter the status of lunch.

The second declares a relative time for both lunch and the meeting. No matter when lunch is, be it now or in three hours, the meeting will occur 15 minutes later than lunch.

'Let's meet 15 minutes after lunch' would make more sense because if you say in 15 minutes that's saying lets meet in 15 minutes and adding after lunch would make no sense at all............ Definitely the second.
