I would apply a self levelling screed to the tiles first, or else the tile joints will show through the vinyl after a short period of time.

If the vinyl is to face heavy traffic, it should be glued down and sealed around the edges with a good silicone sealant to prevent water passing under kitchen units, skirting etc

Backer board is required for wet areas like bath walls. The original plywood placed on the cement slab was probably due to the slab being too new to accept adhesives. It also created a level coordinating with other flooring in the house. You can tile directly over the vinyl. You can tile directly over tile also. With vinyl there's a skim adhesive material you place on top of the vinyl and then mortar over. Full directions in source. Or you can just remove everything down to the slab but then be prepared to remove any residue (hot water, scraper, shop vac)) from the slab base. Then use a product like Flexbond to lay your tiles. .
