Iwo Jima was part of the Island Hopping strategy to get to japan why would we waste nukes on the little island that was just under Japanese influence when we used it on the main island Japan.

Strategically...

They needed Iwo Jima intact to be used as an emergency air field for strategic bombing of Japan.

Even before the island was totally secured they started using it so that heavily damaged planes returning from missions could land there.

We had enough material for three. One was tested in Nevada and the other two were used in Japan. The bombs were nowhere near ready during for fight for Iwo Jima.

One really good reason is, we didn't have bombs. When we bombed Nagasaki, we used the last a-bomb in the world.
