Through the dipstick tube, assumeing automatic.
If it's a stick , there is aplug up on the side of the trans, remove that ,use a transfer gun to inject 90 weight gear lube.

If you got a 66 mustang I would take it to a mechanic and let him do it so I won't jack up my transmission!!

Randy C is right, they make special funnels for that, they have a long skinny spout

you use a funnel and pour it through the dip stick hole that you use to check the tranny fluid. Please don't mix up the oil dip stick and the tranny fluid dip stick!!
