Depends on what you call a city.
If you draw the line at the city limits, Louisville, Kentucky is largest with a population of about 700,000.
If you include the city's metropolitan area, Cincinnati, Ohio is the clear winner at 2.1 million. The City of Cincinnati, however, "only" has a population of 300,000.
And for the sake of completeness, Nashville, Tennessee is the other large city between Memphis and Detroit. It has a population of about 600,000, which is larger than Cincinnati, but a metropolitan area of about 1.2 million, which is smaller than that of both Louisville and Cincinnati.
