For best fit simply double click the column separator line between each column heading. If multiple columns are selected, double click on any separator and all the columns will automatically best fit to the content.

(When you hover your mouse over the line the cursor will change to a double headed arrow cursor - that's how you know when to double click.)

When you say "best fit line" I assume that you mean a regression line that best fits the data. There are many ways to find the equation of this line in Excel. Which you use depends on your needs.

The easiest is to create an XY Scatter chart of your data. Right click the data series in the chart and choose Add Trend Line. If you want to see the equation, then go to the Options tab and tell it to Place the equation on the chart.

Aside from the chart, the next easiest way is to use the regression tool in the Analysis ToolPak (go to Tools --> Data Analysis and choose Regression). If the Analysis ToolPak isn't enabled, you will need to go to Tools --> Add-ins and put a check mark next to Analysis ToolPak.

You can also use the Slope() and Intercept() functions to get the slope and intercept of the line. Finally, there are the LinEst() (linear) and LogEst() (nonlinear) functions. I don't recommend those last two unless you understand how to use array functions.
