CAn I sue for for defamation against my employer, if it was directed at me. Such as an accusal of an act?

It would have to be more than an accusal -- it would have to be an actual (and false) statement that you did it -- and it would have to be a public statement, not one made privately to you or within the normal corporate hierarchy.

There are other requirements for defamation -- but that is the initial threshold.

Yes, you can sue for slander, or libel if it was in writing. You would have to show it was an intent to do harm to you.
