I think it's Comfortably Numb, from Pink Floyd... A great guitar solo.. I play the guitar myself, so i can say that this solo is way more difficult than those shredding solos of our time....
I once read a "100 greatest guitar solos" list, but i really think that "great" in music is something relevant. So it isn't important for me.. Still, there goes the list, just to have an idea...:

It depends on what kind you like.
First of all, in response to the first answer NO JOHN FRUSCIATE! He is not that great, he's good, at best. Flea is the only actually exceptional one in the band, and Frusciate just covers it up with mediocre solos.
Free Bird is a good solo, but it's long and doesn't really go anywhere, it's more of an impressive solo than an actually great solo.
Stairway to Heaven of course sounds great, but Jimmy Page was the oppisite as Lynard Skynard, the solos sounded amazing and were written beautifully, but nothing truely impressive to actually play.
Dimebag Darrell of course has extremely amazing metal solos, but as amazing as he was, his solos rely too much on the same fast patterns and squels, so as amazing as he was his solos aren't my favorite.
Over all I can't pick just one solo, but my favorite soloing works are those of:
Eddie Van Halen (I GET TO SEE HIM FRONT ROW!!)
Stevie Ray Vaughn
Steve Vai
Micheal Angelo Batio
Yngwie Malmsteen
Jimmy Page
Dimebag Darrell

Time by Joe Satriani. It's so haunting, and it sounds like it's in 4/4 all the way through, but I think it's in 6/8 - the tune changes about halfway through and it's spine tingling! =oD
