It means empiracally determining the sequence of a specific gene this is usually done by isolating the gene (often by cloning it into a suitable vector) and then basically using chemical tags that terminate chain elongation at each base type (so a tag that stops A's being added, a tag that stops G's from being added, etc.). This used to be done old scool using radiolabeled and dideoxy-NTPs (they can't elongate) and running 4 different reactions then combining the data. Now we can use fluorescently labeled NTPs and run a sequencing reaction together.
