I don't think this is known. However I will asume that where there are cities, there are roads, which would put you back in the neolithic age. We've found the oldest cities, which were outposts where seeds and agricultural goods were traded that are close to 30,000 old in the Pacific region, and in africa... doubtless there are similarly old cities in other parts of the world.
I would suggest your question just as you asked it may be not the right way to phrase it. It's possible that first there were settlements, then cities connected by roads, and the finally countries. So you see your question may not be precisely ever answerable.
Roads go back, I believe it is safe to say, before even the rise of Sumeria... I don't imagine they were built of stones in those days... we'd probably justlook at them as really long paths... but they connected these seed cities, which makes them the real deal.

The Roman Empire, now know as a part of Italy. When the Romans invaded here in the UK they built roads like the A5 (Holyhead to London).
