It's the rightmost lane at both border crossings.
If you are crossing at San Ysidro take Ave. Padre Kino (parallel to Paseo de los Heroes but on the other side of the canal)
If it's Otay Mesa, take Blvd. Bellas Artes

For the Otay Mesa crossing from Tijuana, the Sentri lane is the lane on the farthest right side as you are approaching the border. It has a separate entrance from the regular lanes, and cement barriers prevent users of the regular lanes from cutting into the Sentri lane.
The San Ysidro crossing point has several Sentri lanes, again in the rightmost lanes, with a separate entrance as you approach the border.
As a rule, the Otay Mesa crossing use to be not as backed up as the San Ysidro crossing, so I usually return to San Diego via Otay Mesa even though I cross from San Diego at San Ysidro. However, I don't think there's as much as an advantage lately as everyone else seems to have "discovered" Otay Mesa crossing (which is east of San Ysidro by a few miles).

There are two sentry lanes, one in otay mesa and the more popular in San Ysidro. To get the San Ysidro lane u need to take Av. Centenario. Av. Centenario is the where Pueblo Amigo is located east of Zona Rio. To get the Otay Mesa Sentry lane which i personally don't recommended, take the Av Bellas Artes and make a right under the bridge that goes to the border crossing and go on the right side of the shopping center. Avoid the Sentry lanes early in the morning, it get real crazy, even veterans have trouble getting in line. One last thing, u need to be a member of the Sentry program to used these lanes.

Avoid TJ, it's not safe or clean and there is really nothing to see there. If you must go find this place, take the Trolley (light rail) from downtown San Diego, walk across, then take a Centro bus into TJ.
