Start with describing the most significant scene or your favorite part.

Here is an example (with a movie):

"Extending his hand to his opponent, young Josh Waitzkin offers a draw to share the victory of the national chess tournament. Jonathan Poe, his competitor, questions Josh’s motives and rejects the offer, still believing his chances to win are high. After twelve quick moves on the chess board, Josh wins the championship and offers a comforting smile to Jonathon. Josh’s many selfless acts are easily detected in the movie Searching for Bobby Fischer. The support Josh receives, his lasting determination, and his kindheartedness are all characteristics that I can find in myself."

I'm afraid that's a rather vague question. It depends on the novel and the essay. Of course, you don't want to start off saying something boring like, "This essay is about (novel) by (author) ..." Perhaps you could open with a quote from the novel that captures its essence and draws the reader immediately into the subject of your essay.

i would use a quote that was [somewhat if not] crucial to the story.

i had to write a critique of the novel Speak, and what I did, was use the quote where the main character got raped. which is the root of the whole story.

or ... you could define the main character(s). ask a question about him/her/them. say something like.. if you were in THIS POSITION, what would you do? then proceed to talk briefly about how the main character(s) handled it.
