For just three weeks it's best to go direct. I always use Emirates (to go home to Perth) as the flights to and from the UK are around 10 hours for each leg. (Perth - Dubai, Dubai - London)
This is much better than the 16 - 6 split you get going to Kuala Lumpur, Singapore, Bangkok or Hong Kong etc.
If it's longer than 3 weeks then round the world is brilliant. I did it when I had two months to travel in and just loved it. Spent a bloody fortune though and had to post masses of stuff home!

If you have only 3 weeks, do not spend any time on stopovers and round the world tickets.
Get the most direct flight and get it over with.
OZ has so many great things to do that you will enjoy every day there, and the time you have at your stopover you will be knackered from the long flight so you would not enjoy that very much anyhow.
In Australia:
The Red center, if you can the overland journey from Perth is very good, but primitive. (Took 7 days, first 3 nights primitive camping in the wild, the last 3 on campsites with swimming pools.)
Take a couple of tours from wherever you are staying, in most towns you get 3 and 7 days tours, some camping, some hotels, some in-between or mixed accommodations.
Do not just stay in the towns, they are nice but the real Australia is outside them.

I went to aus a few years back and didnt stop except very briefly in Singapore for a transfer, BIG mistake! You have to stop somewhere and have a mooch around, in Singapore Airport they offer a free 4hour trip around the city while you wait for your next flight, though i didnt even have time for that i regretted not going. I would stop somewhere in Asia for a few nights for sure.
In aus, if your not a city girl or dont like places like London, avoid spending too much time in Sydney, a couple of days is enough to see all the good stuff without feeling like you do when you go to cramped cities like London. Cairns is beautiful, get internal flights for sure, theyre dead cheap. Also Brisbane is very nice if you want abit of city and abit of the wilderness.

I would recommend a stop over in Singapore, really nice place and very safe. I would suggest at least an overnight
