This kind of question is difficult to answer accurately. When Dirty Harry proclaimed in 1971 that the .44 Magnum was the most powerful handgun in the world his statement was pure hyperbole. The .454 Casull, a far more powerful handgun, had been around since at least 1959. So you see, his statement wasn't all that accurate but it did make good Hollywood hype & advertisement for Smith & Wesson.
Today one could argue that the .500 S & W Magnum is the world's most powerful handgun, then you find out that people are shooting single shot pistols chambered for the .600 Nitro Express cartridge. So you see, that statement isn't accurate either.
But keeping it simple, let's say that it is the .500 S & W Mag. That behemoth revolver would set you back at least a cool thousand bucks. A Taurus Raging Bull in .454 Casull would cost around seven or seven-hundred-fifty bucks. So you see, it won't be cheap.

The Desert Eagle .50AE is the most common powerful semi-auto, whiel the Wildey in specialized calibers can have even more power, though it is a hand built gun and hard to find, and hard to find parts for. I owned a Desert Eagle .50AE, and whiel mine worked flawlessly (except the slide didn't lock back all the time when the gun was empty), I sold it. It's something that seems cool until you ask yourself, what am I going to do with this thing? Revolvers offer a much more compact and efficient package with magnum calibers, though they don't have the same 'movie' impressiveness the semi-autos have. The Desert Ealge also comes in .357 and .44 magnum, and a company called 'Coonan' made a nice .357 auto that was just a bit larger than a standard 1911A1. Expect to pay a premium for one nowadays.

I believe I read an article about a four shot revolver made to handle the .700 Nitro Express, and I know there is a company in Germany or Switzerland that makes 5 shot revolvers in .600 Nitro Express. These are custom or semi-custom revolvers and will cost $10,000.00 or more easily.
Now if you are talking about production revolvers, take a good look at the .500 S&W Magnum, and the .475 Linebaugh Long. Be warned, even hardened and virtually recoil proof handgunners consider the .475 Linebaugh Long to be brutally painful to shoot with full power loads.

actually there are two answers to this question, the most powerfull handgun that is chambered in a handgun cartridge is the 500 S&W. However, there are custom built handguns that are chambered for some very powerful rifle cartridges that make the S&W 500 look like a tinker toy. So hopefully this answered your queistion either way. Good question though.

The 500 Smith and Wesson is the most powerful "revolver". There are single shot pistols built for rifle calibers which can be much more powerful. Both are far more powerful than any semi-automatic.
