The grid doesn't 'amplify' . The number of electrons is simple to calculate. A current of 1 amp for 1 second consistst of avogadro's number of electron charges. Just measure the plate current. You can find Avogadro's number in any reference book. multiply both and then by the time the current flows.

Changing the voltage on the grid will change the current flowing through the tube..... There is a slight correction for leakage and other factors, but they are second order effects. Do the math above.
