Yes, any of those would be a good filling. Store bakeries are starting to use whipped cream as fillings, but vanilla pudding or custard is traditionally used as a filling for Boston Cream Pie. Make a white or yellow layer cake. Split the layer horizontally and fill with the pudding. Frost top only with chocolate frosting. That's a traditional Boston Cream Pie. If your recipe makes 2 layers, you can freeze the 2nd layer until you want to use it (without the filling).

Cook three layers. Make the pudding in TWO batches. One for the cake layers and one for the middle. Both batches should be thicker than usual. Don't make the second batch until just before you need it. Cut the middle out of one layer, leaving only about a 1/2" to 1" ring. Use pudding to put the center layer on the bottom layer. Arrange strawberries in the center of the middle ring. Pour more pudding on top of the strawberries, allowing it to fill in all the spaces. Though you created it thicker than usual, it wil be runny enough to allow you to put it on the strawberries in this fashion. Once set, the putting will be quite thick. Add the top layer, using the pudding again as a filler between layers. The cake will look like a layer cake, but when cut open the strawberries will be a surprise filling.

Yes, if you put the pudding in between the layers it will ooze out the sides, which isn't always a bad thing. (I use pudding in the middle when I make a Boston Cream Pie, and then just frost the top. But I put it together right before it is eaten.) For your cake, I would suggest using Cool Whip (instead of pudding) and then arrange the strawberries on top in a pretty design. Either whole strawberries or sliced.

Yes you can. You can pull out little tubes of cake by poking through each layer with a straw, hold your finger over the end to suction it out, then pour the pudding on to fill in the holes before the pudding sets up, and chill the whole cake. Do this per layer and frost as you like with whatever kind of frosting to cover the holes. Frosting alternatives? Marshmellow creme, whipped cream or cool whip, mousse, powdered sugar glaze, Nutella.
This will work for any flavor of pudding. Mousse as a fluffier texture, might not go into the straw holes as easily...but you can inject it into the cake with a pastry filler, the way you would add jelly to the center of a donut.

You can use anything that's not runny. Make sure you keep your cake in the refrigerator if you use pudding.
