This is not a question we can (or should) answer for you ... WE haven't seen you out riding lately, have we?
If what you're really asking is whether you SHOULD wear underwear beneath your bike shorts (compared with ARE you wearing underwear) ... the answer is NO.
The chamois (the pad in the crotch) of "bike shorts" is designed to soak up sweat and keep you dry, as well as pad the soft bits from excessive bumping around on a bike seat. If you wear underwear between you and it, you aren't going to let it do its job correctly. You'll likely end up with chafing, redness, more soreness from the underwear material rubbing, and overall uncomfortableness.
If you're wearing "ordinary" shorts, though ... it's probably a good idea to wear underwear to give a little extra padding and something else to soak up the sweat from being active.

No you don't wear underwear with your bike shorts. Not only does it interefere with the wicking ability of the spandex, but underwear also has seams in it. These seams create pressure points in places where you really don't want a pressure point.

You do not wear any undergarments with cycling shorts. Cycling shorts are designed to contact the skin directly. The ergonomic pads fit the body and are designed to be soothing when there might be a lot of friction against the saddle.

Jay P and Chris W have the correct answers. If you wish to wear tights in cooler weather -- they go over the shorts.

well, the first thing you have to take into account is the type of person you are. are you a slightly overweight balding man who just bought a specialized allez online for $800 and rides with toe clips? if so, feel free to wear underwear under your $30 black spandex you bought at gart brothers. otherwise the answer is always no. although if you have to ask i'm assuming the worst.

i don't think bikers wear underwear. otherwise you would be able to see it through the spandex shorts they wear.
