Well you can make a picture that is soft fairly sharp providing the file size is large enough. If the picture is just plain blurry then you can only make it less blurry.
The best method is to use photoshop.
Go to Filter>Sharpen>Unsharp mask (It is an old printing term..trust me it works)
Set the threshold to zero, the radius to .3 and the percent to 200%
If you have a large file size you can actually do this a couple of times. (Note it is better to do it in steps than all at once)

Unfortunately, once the picture is taken blurry there's a limit to what you can do to fix it. Stuff you see in movies and on CSI don't really exist yet :)
You can try something like a Sharpen filter in a program like Photoshop or Irfanview, but you'll never get it looking as nice as it would be as an in-focus shot.

I agree with first respondent - the sharpen feature on Photoshop. Maybe you could also draw more attention to the subject which you want to sharpen by delineating it more - darken the rest of the pic or crop just the part you want to use. You could possibly shade around it, but then you run the risk of just completely altering the photo completely.
