We can know whether we are born again by looking at Jesus' answer to Nicodemus:

"I tell you the truth, no one can enter the kingdom of God unless he is born of water and the Spirit. Flesh gives birth to flesh, but ***the Spirit gives birth to spirit.***"

Once you accept Jesus as your Lord and Savior, you receive the gift of the Holy Spirit, who dwells in you and regenerates you. Here are the supporting verses:

"And you also were included in Christ when you heard the word of truth, the gospel of your salvation. Having believed, you were marked in him with a seal, the promised Holy Spirit, who is a deposit guaranteeing our inheritance until the redemption of those who are God's possession—to the praise of his glory." -Ephesians 1:13-14

"And if the Spirit of him who raised Jesus from the dead is living in you, he who raised Christ from the dead will also give life to your mortal bodies through his Spirit, who lives in you." -Romans 8:11

Also, as a previous poster mentioned, after you've been reborn, you should have a changed life - mind you, this doesn't neccesarily happen overnight; God is patient with us. My entire attitude and perception of life changed also. I've realized that true satisfaction in life is not possible if I strive only to please myself - true fulfillment comes from striving to please God, to bring Him glory, and to shine the light of Christ's love to those who are hurting and lost in the darkness.

I know the Holy Spirit lives inside me because He convicts me of sin, which hinders my relationship with God. He gives me the power to defy my own nature by giving me the strength to turn from my sins, the compassion to care for people I don't neccesarily feel like caring about, and the heart to forgive and pray for those who have mistreated me and never asked for forgiveness. He comforts me and has, in the past, filled me with the "peace that surpasses understanding." He's given me the words to say when speaking to a close friend about salvation. He's guided my words and actions during mission trips. Sadly, at times, I've grieved the Holy Spirit (Ephesians 4:30-32). Believe me, you'll know if you've done this - it's the most awful feeling in the world. He's prompted me to pray for people I didn't even know but who needed prayer.

Basically, you'll know if the Holy Spirit lives inside you because He will guide and teach you. If you are unsure personally whether or not you're saved, ask God to reveal the truth to you and He will.

You know you are born again when you accept Jesus as your Lord and Savior.
Ask Him to forgive you of your sins and repent (turn away from sin).

Jesus will send the Holy Spirit who reveals Truth.
You become a new creation in Christ.

You begin to change the way you think, act, speak,etc.
You start becoming more like Jesus.

Born again for me has meant a whole new life. Everything has been transformed. My attitude, perception, interests, desires,goals, ideas ..all have changed since being born again. Before my faith was no more important than twice a year forced visits to church. Today I am a devout Catholic and God did it all by introducing himself to me and calling me to try a new life. One with Him. And with His plan at the helm. If you've been born again there no mystery about it having happened.
It was transforming like a near death experience.

His name is Jesus Christ and every word he speaks is true.

Yes, and remember what Jesus told him? Ye must be born of water and the Spirit - in other words, baptized. That's how I know I am born again. I have been reborn through water and the Spirit, just as Jesus said.

Baptism is spiritual rebirth, through which we receive sanctifying grace, and become children of God and heirs of heaven. It's tragic that it has been reduced to a mere symbolic gesture in manmade denominational religion.
