Timing is essential in being a good comedian. You can have great material, but if you don't time the punchlines right people will think it's the least funny stuff they have ever heard. I'd like to point out some examples.
Have you seen the cell phone service commercial with the van that pops out? The guy says "Thinking about leaving us? Well I wouldn't if you ever want to see fluffy again!" The lady says (without any pause or anything) "But that's not my dog!" This had such great potential. But without the proper timing, this commercial was absolutely stupid, and was crap. With some comedic timing, it would have been great.
I would suggest practicing timing for your jokes well before a show. Give the jokes to friends. If there immediate reaction isn't laughter, it's probably not funny. People shouldn't be goaded to laugh at a joke, it should just be funny.
Another important thing is voice inflection. Make sure and practice not only when you say something, but HOW you say it. Giving a slight raise in pitch or slight accent can make a great difference in wether or not something is funny. Some comedians are natural at timing and voice inflections, just like some musicians are natural musicians and don't need quite as much practice. Others need to practice, but are just as good when they have practiced.
