if you're a beer fan I would recommend Stout. They have a great selection and its a block away from Penn Station, doesn't get much more midtown than that.

There are over zillion places to go for happy hour in Manhattan. It all depends on your mood, location and the crowd you want. But if you want to enjoy the Manhattan view and also the drinks, check out Hotel Pennsylvania Happy Hour. It's located on the top floor and excellent drinks. Of course a bit pricey.
