Of course! Any time a director gives you their undivided attention it is an excellent sign that you are progressing as an artist. Just remember that no matter what happens, you need to get something out of the experience as well. Don't be afraid to some informal critique if the opportunity presents itself. That's a good thing, almost as good as a "call-back", or secondary audition. It means the director doesn't think he would be wasting his time by seeing more of what you can do.

It means that they want you to perform with them. If you want the job, it is a very good thing. If you are wondering why they don't just take the practice as a "real" audition: It is about avoiding preferential tretment and more importantly, looking like they are avoiding preferential treatment.
