25mg. If there are .5g of codeine in 100ml of cough syrup, then there are 500mg in 100ml of cough syrup - so 5mg/ml. A US teaspoon is approximately 5ml. So 5ml x 5mg/ml = 25mg.

Even though the amount isn't exact, a teaspoon is generally accepted as 5 ml.
This is not a house hold teaspoon by the way.
