That will depend on which country issued your passport. It has nothing to do with the B2 visa as Canada will not recognize that.

If you have a passport from an EU country, then you won't need a visa to enter Canada. However, if you have a passport from say Algeria or Russia then you will need to obtain a visa first.

Check with your nearest Canadian embassy or consulate, if you need a visa it will usually be a very similar process to getting the US-issued visa.

Canada customs will not care about where you are or what visa the US gave you. What matters is your citizenship. As you didn't include your citizenship, here is the list of who need visas to enter Canada. Also, if you plan on going back to the US afterwords, double check to see if your US visa allows multiple entries.
