B&H Photo on Ninth Avenue and 33rd Street - huge selection, great prices, great service. They have a wonderful reputation all over the country. Note that they are closed Saturdays and open on Sundays.

Hint on negotiating - let them sell you on a camera, don't act overly interested, but let them continue the sales pitch. Maybe buy an inexpensive accessory or something. Then try and walk away, the price will drop rapidly to make the sale. I was in a store on 7th Ave recently, they were trying to sell me a lens, price started at $850, by the time I was walking out the price dropped to $250 and they threw in a polarizing filter.
