I have just yesterday returned from a two week self drive tour of Tasmania... :-)
Take the ferry across if you're already in Australia..worth the experience. You might want to book a cabin, but sleeping in the 'business seats' wasn't too bad. I did it with 4 kids and they were great.
If you take the ferry option you'll land in Devonport...you can drive around the whole of Tasmania in two days..it's that small, but of course you won't be doing that!
What we did was, we went from Devonport, stayed in Ulverston and took a day trip up to Stanley (northern coast). Then basically took the road all around the east coast and then back up the west coast and also made day trips up the middle.
1. The east coast is fabulous.
2. From Launceston head up the Tamar Valley, lots of vineyards and history along the way.
3. Head across to Scottsdale and along through the old mining towns. I fell in love with a place called Derby which is an old tin mining town, totally recreated and is just wonderfully picturesque.
4. We stayed in St Helens - fishing town, lovely spot,. You can pretty much go to the visitor centres as you find them - they are able to book you into accomodation and trips at no charge.
5. Freycinet National Park is wonderful.
6. MUST SEE PORT ARTHUR - I would go back to Tasmania just to go there again. Bountiful with really interesting convict history. Great day trips in the area.
7. You must also pay a visit to the CADBURY FACTORY in Hobart - if you love Chocolate it's worth the tour and certainly worth buying ALL that chocolate at criminally low prices from the factory! I left with bags/boxes of chocolate.
8. Huon Trail is interesting if you're into the natural resources of Tasmania...my kids weren't that interested so I skipped a good portion of that trip (south west of Hobart).
9. The west coast I found incredibly boring - the landscape is vastly different from any other part of Tasmania..has it's own kind of beauty, but really it's just windy roads with nothing interesting to look at.
10..that said, if you're driving, you need to take this route to get up to Strahan. There is NOTHING worth stopping for in Queenstown, Strahan is very pretty and relies heavily on tourism and fishing industry to keep it alive. There is a lot of indulgent accomodation and restaurants in strahan...and it's worth booking in to see the Pirate Theatre at the Information Centre. Recounts a true story of mutineers on the Frederick.
11. Cradle Mountain is worth staying at if you love the outdoors and day walks in the alpine region. I'd go back to stay there.
12. Certainly go to MOLE CREEK to have a look at MARAKOOPA and KING SOLOMON'S CAVES.
Do NOT visit TROUWANNA WILDLIFE PARK..probably the greatest waste of money yet.
It's not hard to get off the beaten track in Tasmania..there really are no highways to speak of. The pace is a lot slower, it's a lot like New Zealand and well worth a visit. Compared to the rest of Australia, Tasmania would be my favourite state.

Tassie is so damn small you can see mostly everything in a couple of weeks. Launceston to Hobart is only 2 hours on the highway. I recommend the Port Arthur night tour, that was great. Read "For the Term of his Natural Life" before you go, then you'll be able to see the places in it. It's always cool to see places you've read about. Eaglehawk Neck is really as thin as they said in the book! Also, do a half day cruise at Strahan.

Tamar Valley, near Launceston, for wine (until they build the pulp mill)
Freycinet Peninsula (if money is no problem, stay in Freycinet Lodge.
Tasman Peninsula.
Hobart.
Lake St Clair.
Strahan. Do a cruise up the Gordon River and a flight over the Franklin.
Cradle Mountain.
Burnie cheese factory.
But pretty much anywhere - the place is beautiful

I've driven round Tasmania twice, and had a great time without any must see/do. We tended to just see what came along next and it was all pretty good, except for the taste of the water in Queenstown. Hope you have fun!

We hired a CAMPERVAN and travelled all around Tassie.....it was the best vacation ever. You are so totally independent. The tourist bureau has tons of things to see and do.....and if you just keep on drivingaround the coast and up the middle you are going to see nearly everything. You will get sidetracked so often....but it sure is the experience of a lifetime.HAVE FUN!! Also when you get off the boat or the plane there are tourism reps there handing out bags of booklets of everything to see and do...we really appreciated that.
