If you go to church and drop $500 in the bucket, with no receipt, you won't get through the audit, if you have one.

If you give $500 to your college, but ask that it be kept confidential and anonymous, they employee in the "alumni giving office" can give you a receipt, but not tell the rest of the world who it is from. In this case, you have a receipt, and the IRS auditor would be okay with it.

No, because you must get a receipt from the organization in order to be allowed to deduct it, and you couldn't do that if you made the donation anonymously.
