you can make them several days before. you can make about 20 shots depending on how full you fill them.

You should make them about a day before so they can set up, and go easy on the vodka, or whatever you are using. You want people to taste the fruity jello, not grain alcohol.
To answer your question, depending on the size cups, you can make 20 shots or so. Have fun!

I would make them the day before.
We have made them with a variety of different liquors. A couple of good combos are orange jello with peachtree schnapps, watermelon jello with midori. For the rest I use vodka.
Oh and the amount of shots depends on the size of your cups.
