I know you've got several answers already, here's my two cents:
"Espacio" means "space".
But... it can mean "room" when you talk about a extent of space occupied: "There's no room/space for this" = "no hay espacio para esto".
It's also when used talking about a 'period of time' and the 'outer space.' = "Espacio de tiempo" and "el espacio exterior"
"Room", when meaning a part of a building is "habitación", "cuarto", "pieza" = "this house has three rooms" = "esta casa tiene tres habitaciones".
"Cuarto" in other contexts means "fourth" and "quarter".
"Block" as in "it's two blocks from here" is "cuadra" = "Está a dos cuadras de aquí".
"Block" as in "to go around the block" is "manzana".. Yes, like the fruit!!! = "Darle la vuelta a la manzana".
"Block" as in a piece of material is "bloque" = "a block of cement" = "un bloque de cemento".

Espacio is room in the sense of space.
For example: No hay bastante espacio= There
isn't enough room.
Cuarto is a room you inhabit.
For example: Quisiéramos desocupar el cuarto.
We'd like to check out of our room.
(Useful phrase if you're on a trip!)
Block, in the sense of city block, is manzana.
(Yes, the same word means apple!)

Examples:
