You sound like a lady with principles. Good for you. Since those principles include something like "It is a sin to tell a lie.", stick to your principles. Tell the interviewer the truth.
It may cost you the job, but sticking to your principles will help you hold your head higher than lying.
There are times principles have a cost. This may be one of them.
Something will come up. I am currently looking two jobs that only works weekdays. They do exist.
I've had jobs that I traded days with a coworker. He was 7th Day Adventist. I worked his Saturdays and he worked my Sundays. It's called teamwork.

If you can't work Saturdays and they need you to work Saturdays, then the job's not right for you. Pretending it is right by lying to either your employers or yourself may help you out in the short term -- I.E, you may end up getting the job -- but it will hurt you when you have to either let them down by not keeping your work schedule or let yourself down by not meeting your personal commitments.
With some jobs, you can negotiate such things. For example, you could offer to work Friday nights but not Saturdays. However, if they really need the Saturday help then trying to negotiate that point with them will probably frustrate both sides.
If the person helping you to get the job is ignoring your personal needs or telling you to do so, maybe you need to be more assertive in letting her know your limits -- or, maybe she's not the kind of person that you want helping you.

How do you feel about losing your religion for a job? If it doesn't make you feel good, don't do it!! You may be ok for a few months, but you will be mad at yourself down the road.
Technically, businesses aren't supposed to base hiring you on your religious beliefs, but if they really need you on Saturday and you really can't work then, then the decision is all on you.

I guess this is what you call "TRIAL"
Time for you to make a choice. Doesn't sound to me like you are going to have a tough time making it either. After all, in your own words, "I am not willing to give up my religious beliefs just for a job" Is this a question at all, or are you just looking for justification for what you have already decided?
