I see the range of opinions ranges a lot on this question but if you asked a more specific question, you may have some better responses.
Many prefer the 9mm as it is a very popular round, worldwide. They usually have fairly large capacity magazines and in the right loading are fairly effective.
The .38 is also a good round but with the other options for calibers out there, you could do better.
Of course your choice is your own, for whatever reason you may choose.
I prefer a .45ACP. The stopping power of the .45 is unquestionable at combat handgun distances, and the variety of ammo available is very wide. This is also true for the 9mm and .38, but, I still prefer the .45.
It is no secret that the 9mm is the most popular caliber throughout the world, but the US military and law enforcement communities are migrating back the the .45 due to it's sheer stopping power/effectiveness.
The 9mm had it's turn in the limelight, but after about 20 years of data, the military and the LE communities are finding that the effectiveness of the 9mm is not as great as many hoped it would be. They (the 9mm's)are here to stay but, the .45ACP is creeping back into the holsters of Marines, Sailors, special operations forces, and Police officers, even as we type. I think the reason is that after trying the 9mm and .40S&W for the past few decades, the .45 is unmatched when it comes to an auto-pistol round.
Why don't you consider the .40S&W?
It's right above the 9mm, and right under the .45. Most of the pistols (various brands/modles) handle pretty well considering their compact sizes, and ammo is reasonable.

Depends on what you are comfortable with. I have a large frame and can handle follow up shots with a 45 with ease. My fiancee cannot recover as quickly when shooting the 45, but with the 9mm, she can double tap and triple tap within 4" circles.
It basically comes down to what is the biggest caliber you can comfortably shoot. For me, its the 45acp. But every collection needs all of the above. I have 10 or so 38/357's, 3 9mm's and maybe a dozen or more 45's.
For carry with heavier clothes, I have a Glock 36 45acp, for carrying with my suit on, its my S&W 342PD 38 spl and for range carry, its my CZ SP01 9mm for the ability to throw 19 rounds down range with deadly accuracy and without breaking the bank.

No on 38 special, it is the weakest of the bunch. Most Americans prefer 45, they'll claim that 9mm doesn't have the stopping power to match a 45. However due to the fact that 9mm is the most popular pistol round on the planet (most produced) and most armies around the world use it. If 9mm was a bad combat round don't you think everyone else would have abandoned it by now for the 45.? While 9mm is weaker than a 45. it is still more than up to the task. My compact Glock holds 15+1 rounds of 147 grain 9mm hollowpoints. The 1911 (45.) holds 8+1 that's a big difference

What for? The .38 Special is very versitile and fair mild to shoot. You can load snake shot in it if you live where you might have to shoot snakes. It is more user friendl and requires less attention than your auto loaders.
Of the three I'd go with the nine or .45 strictly for defensive purposes. The nine generally holds more ammo than the .45 and with the hotter rounds it is as good as the .45 loaded with ball ammo. If you just prefer a wheelgun then the .38 will work with the right defensive ammo like from Federal.
Best.

A tie between .45 and 9mm. I just don't want or need a .38/.357 wheelgun anymore. I still love them (especially 6" standard "non-lugged"/classic barrels) but I only buy what I shoot. I have a 1911 series but toy with the idea of getting a 9mm "classic" (all-steel model).

.45 acp hands down. The 9mm is just a glorified .38. Something to think about.. If a 9mm couldn't even kill the Pope, why would you carry one?

The .38 special is the best. It is by far the most accurate of the 3 rounds.
