In Prophecies you get the first signet from a quest in Lion's Arch. There is a man on a hill near the North and East exists of town that will give you a quest. After you complete the quest he will give you a signet and another quest to use it. After you use it and complete the quest, he will give you one more.

Each signet only works successfully once. Kill a boss (anything with an aura or that gives a moral bonus) then use the signet while standing near the corpse. The boss' skill list will open. Click on the skill you want and it will IMMEDIATELY replace your signet on your skill list. If you choose to cancel instead, you keep the signet. You can capture any skill this way, but you get an exp bonus for elite captures.

After you have gotten the very first signet you can buy more from skill trainers for one skill point and whatever your current skill cost in gold is. You do not spend another skill point when you capture a skill.

You get cap sigs from skill trainers after you reach a certain point in the game. You can buy as many as you can afford and use them to cap Elite skills from bosses.

In Prophecies, you can take a quest from Magi Malaquire in Lion's Arch to get a free cap sig. Afterwards you can buy more sigs from him. You won't start finding Elite skills though until you reach the Crystal Desert missions.
