You didn't say how old these kittens are, but--if you haven't seen this one for 2 days, I'd say your dog killed it--probably not intentionally, the dog was playing with the kitten, but the kitten was too tiny to take it or to get away from your dog.

How do you know that the kitten hasn't been drinking if it is gone? If the kitten is for sure gone and you were talking about the dog not drinking, take the dog to the vet and I suspect the dog ate the kitten. You need to add some details.
