Depends on the fish and what kind of hooks your useing.
For some saltwater fish, I tell customers to not set the hook, and just leave it alone until you see the rod bend down. Ive seen TONS of people miss fish because they try to set the hook when you don't need to. Like shark fishing, alot of people think you should set the hook the second the fish takes it, but instead you need to wait a few minutes and let the shark swim with it and swallow BEFORE you set the hook, otherwise he might spit it back out midfight without being hooked.
If your using circle hooks, there is no need to set the hook with them, they will set themselves. They are designed to set themselves when a fish tries to swim off with them.

Once you get a nibble, let the fish play with it a little. Once he/she really starts to tug, set the hook. Just like the first poster said.
Sometimes baby fish do a lot of nibbling and never really take the bait. Be a responsible fisherman and don't try and rip the jaws off a fish.
If you know about fishing, you know what I mean.

Depends on the fish. Some fish are nibblers, others are whackers. With nibblers, you have to wait for a slight tug, then quickly lift up the rod tip about a foot. With whackers, they hook themselves and no need to set the hook.

well it somewhat depends on what your fishing and what your fishing for, but usually a hard jerk to the side in a upward motion works fine.
