I am wanting to put a two inch lift block in the rear of a 2005 dodge ram 1500 4x4 truck. My friend is telling me that this will mess with my rear axle and cause major damage. I've never heard of this but maybe someone can help me with this.

Won't hurt the rear end, but the u-joint angle may be off. It can be corrected by shims, in the case of square blocks, or by blocks that are thinner on one end.

it wont hurt the rear end but it will really put the drive shaft in a bind on it,you,d need to re-set the angle on the rear end for this to work right ,good luck on it.
